Google home tissue culture: you should find what you need. This is the recipe I use at school:
1.1-2.2g/L Murashige and Skoog (MS) salts
10.0ml/L Gamborg B5 vitamins 100x stock
20.0-30.0g/L Table sugar
6.0-8.0g/L Agar
1.0L Deionized water or distilled water
Dissolve everything in 900mL water. Adjust pH to 5.5-5.7. Autoclave at 121*C for 20min.
I haven't looked into this yet, but off the top of my head, to do TC at home:
- use a complete fertilizer to replace the MS salts
- use a B12 vitamin tablet instead of the vitamin stock
- agar can be bought from Asian supermarkets
- adjust pH using vinegar and dissolved baking soda
- use litmus paper instead of a pH meter to determine pH
- dissolve everything and aliquot into glass jars and lids before sterilizing
- use a pressure cooker instead of an autoclave
EDIT: bring volume up to 1.0L after pHing
