Hi has anyone in here have tried home media preparation? I'm curious to know, not only if it works, but if it works well... I had only found two distinc recipie (also found 2 others, but they are kind of inspired from one of these two...) www.une.edu.au/agss/hort/horticultural-science.php#item6www.aquabotanic.com/stiffaquatic.htmif not, if considering buying Murashiche Skoog (MS)...I've read that tom use the normal one, but has someone tried the other ones? they're about the same price, and I was curious about their results. about the hormones...they're plenty!!! Wich auxin // cytokinine do you use? And wich giberilline? I guest you go for the cheaper, but I've been told that some plant may required specific hormones... also, do you know of a place where we can buy cheap balances that can measures mg...I only know of ebay, but a little worried about being had as I've never used that system... finaly, last question, where did you guys buy the phmeter? I went to the petshop, they only have the liquid thing. by the way, I already have the ph strips...it doesn't work for me...lol...(I not colour-blinded!!!)

wow...for the balance, I'm gonna have lots of reading after my final exams...lol I'm ''haste'' (thats' how we say?...exited about it^^) thanks (by the way, if anyone know of cheaper place for hormones or MS or whatever, keep posting...we all want to know...lol ;D)

Cheapest hormones is to ask to see if anyone here has any surplus to sell or trade. Phytotech is the cheapest for most things. If you are ordering enough the US$15 (US postal service not UPS) isn't too bad.

thanks^^ but unless you have a lot, I'd rather buy directly from phytotech (unless I find another company cheaper...or a chinese friend...lol...lots of thing are made in china these day!!!). I prefer that way as I would buy big amount to be settled once on. then again, I'll first have to read the article on the balance to see if I can do it myself. after that, phmeter and finally...tissue culture!!! actually, for the phmeter, I've found that we could measure it with a voltmeter... pH=-17,2*V +7the only problem is that it needs to measure very small number...pH 4 is about 0,174 volts, pH 7,2 -0,012 and pH 10 -0,174...so, now, I'm trying to figure out if they're something verry common that we all know but just didn't think about it...

The problem with hormones is you buy 100 mills. You usually use 1 to 10 drops/ 500 mill of medium. Unless you have a full scale TC business the hormones will go bad before you can use them up. Get a real meter. There is a temp factor as well. I think I paid about 80$ for mine they are handy. I don't usually adjust the PH much. When I mix a batch up it is usually around 6 and it will increase a couple of points if I let it sit for a couple of days before adjusting and sterilizing. For some reason the PH drops to 3 in my medium for VFT after a month or so, I still can't figure that one out. You are diving into a big one with the TC it is rewarding but can be time consuming. If you research carefully what you want to do and what you "need" you can make an order to Phytotech for around US$50 or so with postage. Make the order through email asking for USPS (US Post Office) option before paying. Postage will be ~US$15 which isn't too bad if you order everything you need. Do not use UPS as they will charge you more than the order costs to move it through customs. You won't know about the extra UPS charges until delivery.
I've found Phytotech is cheapest except for Gibberellic Acid (RareExoticSeeds), food grade agar (bulkfoods.com), Activated charcoal (Big Al's in USA if you have a USA contact), and potassium citrate from jdphotochem.com and citric acid from the grocery store.
You can mix your own 1N HCL from muriatic acid at the hardware store (only if you are super-very careful) and 1N NaOH from lye at Home Hardware (also potentially very dangerous especially to the eyes!!!). I have to say don't do this as you can potentially burn yourself severely or blind yourself mixing the solutions. You need these to dissolve hormones. The 1N solutions are less dangerous than the strong acid and solid lye.
Almost 100% Ethanol and isopropanol also hydrogen peroxide are available at Shoppers Drug mart. Bleach of course from anywhere.
As for the pH meter, you will need a pH electrode which is the more expensive part of the pH meter so there is not much point in using a voltmeter unless you are making your own pH electrode (not an easy task!).
The homemade scale does need a voltmeter that measures down to 200 mV. I checked out 1 mg scales online and they were in the hundreds of $. You could always buy a 10 mg scale for maybe $30+ or so and just make a stronger stock solution and dilute it. You will need reference weights too, at least one (I bought a 10mg one.)
