Thanks for the offer lloyd. I will try a health store or two this weekend as well as an Asian grocer in the area. If I cannot find it, I may take you up on your offer.
I kind of figured the 30mL would last forever, but it seems like it would make things a bit easier to do at home. I sent them off an e-mail now, so we'll see what they say.
Also seems like the NAA wll last forever since the lowest quantity phytotech offers it is at 25 grams, still looking around before I commit to ordering that though.
Cheers

Don't order NAA, I can give you some for free as I have enough. I can also "lend" you amounts of other stuff if you aren't ready for a phytotech order. So pm me before ordering and I'll tell you what I can help you with.

Perfect, thanks again lloyd.
Also, got a reply back from ppm for tc, they said the shipping is included in US, but since it was my first order she would waive the shipping cost. So, I've sent my order in for that. Thanks lloyd, you've been extremely helpful!

I guess a bit of an update,
I got my phytotech order in, just some basic things for now -some MS media, lids, and BAP.
They sent the wrong lids, but were excellent in correcting the order, the right ones should arrive within a couple days. I couldn't resist trying to make some of the media, so I used the modified mason jar lids for the first few. I put a VFT flower stalk in one, and also some VFT seeds, and Ping seeds in another couple jars. No BAP yet, as I still don't have any KOH to dissolve it into.
So far no contamination... its been only 5 days though!

Very nice renesis, you don't need the BAP for seeds. It's only used on explants to promote shoot proliferation. Keep us updated!

So, I got home today and the flower stalk on the typical VFT I had decided to shoot for the top of the terrarium while I was gone. I thought I could wait for the weekend before taking it down and putting into some jars, but it must have gone from 1.5" yesterday to a bit over 3" today!
So, I sterilized it, cut it up into 3 pieces and put each into a jar.
As I understand it, BAP isn't required for tissue as well unless you want plenty of offshoots, correct? Otherwise the flower stocks I've used are a waste. Hopefully at least one of them works!
Also, the mason jars I'm using are really annoying, the walls have decorations of fruits and make it really difficult to see whats happening inside.
Will definitely post some updates once I see any changes!

Little bit of an update:
I've source KOH and at a good price. I found I needed some to clear out roots to search for endomycorrhiza in some plants at work as well so I added it into the lab order. Worked out to $9.80/100g.
The first flower stock has succumbed to contamination- appears it came out of the bulb area. Do you guys cut this part off? The other three I think have been over sterilized, they're black at the ends and there's hardly much life in them. The VFT seeds haven't done anything yet... must have got old ones (ebay).
Still on the lookout for clear undecorated mason jars if anyone has seen any!

I have NaOH if you want some. Or: www.jdphotochem.com/44- Potassium Hydroxide $5.00/100g is the cheapest place I have found and shipping is also good as it is Canadian. The article on ICPS says to throw away the top part of the VFT stalk as it is too iffy to "sterilize". I rinsed the stalks in distilled water with Tween 20 before decontamination. I eventually settled on a solution of No-Damp 0.1cc, 0.4cc PPM, MS 43 mg in 10cc distilled water for 4 hours. Do not rinse, plate on medium with PPM. With this method I had no contamination but death occurred by phenolic oxidation. I think the hormones I used were too harsh, next time I will use no hormones or maybe 1/10 of what the ICPS article called for.

Wow! Nice prices on a lot of that stuff. I order most of my supplies for work from either prolabscientific.com in Montreal, or will pop in to efston science in North York for small things. The company is footing the bill on the KOH, and it won't be a big deal if I borrowed some for personal use. In fact, I did quite a bit of the prep work for my first batch at work using the balance and pH meters.
The contaminated flower stock is fully black now. I don't know why I'm still hanging onto it.. The other 3 so far haven't shown any growth, but the tissue is still green so I've still got hope.
I will try your method for sterilization and see how that works out for me! Thanks lloyd.

absinthe: Right on! Haha, that would be my 'expensive' hobby.
blokeman: I got my first set of mason jars from Canadian tire, but the only clear glass ones were 500mL and above, all the 250mL and below had the decorative glass style. Maybe I'll try another Canadian tire store...

What I did was order some phytocaps from Phytotechlab and bought some baby food jars from the store. Empty the jars and use them. Still cheaper than buying mason jars. Or hunt fora family with a baby, ask them for baby food jars.
