So, I've been reading a lot about TC in the past few days and decided that I'm intrigued enough and would like to give it a shot.
Especially since I have some access to certain equipment at work including a digital balance scale, pH/E.C. meters, and various glassware.
So, from my research, I believe I would need the following:
- Plastic Box to work aseptically in - MS Salts w/ NPK and micro's - Growth Hormones - Which specifically should I be after? - Autoclave caps for baby food jars - Which from phytotech works best? - Baby Food Jars - Can I use the small mason jars for canning? Will the same lids work? - Agar - Can I use the cheap bulk stuff from say bulk barn or a health food store? - Forceps, Scalpel, (I have these) - Isopropyl alcohol, hydrogen peroxide - figuring Shoppers for these - Bleach (10%) - Pressure Cooker - Check - PPM - Alcohol lamp
Is this good to get me started? I'm in microbiology by trade, so I know aseptic technique pretty well.
Anything else I'm missing? Or any tips? I've pretty much read through the forum topics here and on a couple other sites.

edit- almost forgot, I'd like to start off with some Pings, Drosera, and VFT!

I am also a newbie with TC, I have learned a lot from Lloyd so maybe he can answer your questions. I am still not worthy hehe.

Agar: yes you can buy/use the one in health store, i've done it for years.
Mason jars: as above, but lids can/should be adapted for some ventilations, such as drilling a hole through them and glue some 3M water resistand 'band-aid', or somehygrophobic cotton ball. Using them laid on their side allow you to have a larger area of media, and better light penetration.
Hormones: some on the basics one: BAP, KINETIN, IBA,and NAA, plus perhaps some GA3 if you want to take some seeds out of dormancy, and/or biocide (PPM, etc) to help you have a control on the contaminations.

Nice Tom, I was reading the early posts in the TC section and found that you are one of the pioneers here in the forum with regards to TC. I thought you stopped doing TC, my bad. Laying the bottles to their side is an awesome idea. Thanks Tom for the info.

i did stop doing TC and dont think doing some before a few months or years Mason are great, but some lid customizing are to be one since the lid and ring dont stay together when you open the flasks, which lead to some bad manipulations, accidents, increasing contaminations risks.

I believe that Phytotech carries lids that are compatible with some mason jars. I have to check on the size.

Great info guys.
Are the mason jars okay in the pressure cooker? Even with the metal lids? I don't want any explosions or anything since I believe the other type of lids allow breathing when autoclaved. As long as the modification with the water-proof band aid works, I'll probably go with that.

Yes they can, they are made for this, whe you are canning low-acidity/meat food. Use as described in this link www.homecanning.com/can/ALStepbyStep.asp?ST=6They wont explode if close 'properly' (see link above) and there is no risk if you punch a hole in the lids (and cover it with coton/adhesive bandages), as hot air will be able to go through it (but i can't assume any responsability with accidents!). Be careful on how to unload the pressure cooker: a cool wind/drastic air change around a very hot glass jar can lead cracking/explodig jar. I usually let the presure cooker sit a little while unopened when the pressure got to 0, so the jars cool a little before i lay themon the side for the gelification of the media in the good orientation.

That makes a lot more sense now. I've got some GBA already, need to buy some PPM somewhere. I'll get the hormones and MS from phytotech.
Are there any good sources on the effects of different hormones on CPs? Or even just generally?

I'm in Toronto, so if I can help you out, I will. I like babyfood jars and the C070 phytocaps from Phytotechlabs. I have way too much of certain stuff, like Casein and NAA. Phytotechlabs is good but always ask for USPS, never UPS. UPS rips you off and with USPS you avoid taxes, duty and brokerage. You really don't need any PGR's to begin with. If you're ever in Toronto, I can show you my (dinky) set-up.

tom, thanks for the links. Looks like I should pull out my Biology of Plants Text, I remember working with some hormones back in first year.
lloyd, that would be great! I'd love to see it. I'm not downtown too often (save tonight for the Leaf game), but I will keep you in mind next time I plan on going down there. Thanks for the USPS tip, I have enough experience with UPS ordering parts for my car from the US. I don't think I can even count all the times my packages have either been lost, smashed, or charged ridiculous brokerage fees.

However, these are getting increasingly hard to find. I had to look many places before I was able to find the standard mason jars. Everyone is selling the wide mouth ones.

Perfect, I picked up a set of the standard mouth mason jars today. I will order a set or two of those lids.
I am having trouble finding Agar though. Does anyone know where its available in the GTA? Bulk Barn was a no go.

One more thing... Is there a better (cheaper) source for PPM than www.ppm4plant-tc.com?I'm getting a price of $44US for 30mL shipped, that's $30 for the ppm and $14 for the cheapest shipping option.
