I was reading through this thread: ocps.proboards78.com/index.cgi?board=tissueculture&action=display&thread=2096and though of a couple of questions that I'd like to ask the people that have TC experience: 1) Where to you keep the flame that you use to sterilize your instruments? I am confused about whether or not it's under the hood (or plastic container like some people use and that I will be using). If it's under the plastic container, is it safe to mist the inside of the container with alcohol while the flame is going? Sure seems like it wouldn't be. I'm planning on using my camping stove to clean the instruments. I've read before that this works but I'm afraid of spraying alcohol in the same space. 2) How likely is contamination AFTER the baby food jars are sealed? Is it necessary to place the jars with the plant tissue and media someplace where it is very clean? I was hoping to put them in my terrarium where I have some other seedlings growing. Would that pose a contamination problem? Do most of you use medical tape or florist's tape to seal the lid better? I've read that this is important, but I haven't seen a good photo, so I am just guessing that you apply the tape around the edge of the lid and overlap it on the baby jar. Is that right?

Thanks Hackerberry. I've scoured everything on those websites and I think I have pretty well digested it all. Those questions I had were after I've read those pages, though it's possible that I missed some information when I read them and my questions were answered on those sites.

Mist with alcohol and wait for it to dissipate before any flames. Careful with flames inside plastic, you could melt the box. I flame my instruments outside the box and then leave them in 99% ethanol in the box for safety reasons. Don't bother with film on the baby food jars-I have lots of extra caps designed for TC for baby food containers. PM me and I'll send you some, free except for postage. I put my bottles with the caps on inside zip-lock baggies to keep mites out. If it's a relatively clean area you could just use the caps alone.

Thanks for the reply lloyd. That clears up my questions! I guess that putting the bottles inside zip-lock bags is ok? They don't need to breath?

I leave mine in Zip-lock bags for months with no problems. I doubt they are really super sealed, probably permeable to oxygen.
