I'd have thought that 4% PPM would be toxic if you left it in contact with the plants.

Hi lloyd,
My plants inside the tc bottle is still growing and the plants are the gardenias. I'm pretty sure I used 4% PPM. Those are the ones that I'm going to replate soon along with the vfts.

What's the approximate budget to get set up for TC? I've seen a "Kitchen Counter" kit online that sells for about $250 US!!

I don't have that kit. I sort of made my own.
It can add up as you upgrade your setup. For instance you can start with pH strips and then buy a meter; or start with a glove-box and then build a laminar flow hood.
If you make an order from Phytotech(US$) (USPS is safer and around $20) get a few 1L MS foil packs ($3), a bottle of agar ($19) and some Phytocaps ($5). With postage that would be ~$40-$50. You could use food grade agar or just cotton balls to save money. Baby food bottles can be scrounged. PPM is nice and costs US$30 with shipping for 30 ml that lasts a long time.
pH strips are a few $. A small scale can be bought for about $20 or so. A plastic bin works as a glove-box. Sterilizing can be done with a microwave or pressure cooker ($50). A few pieces of glassware, cheap forceps, little spoons.
Odds and ends like bleach, alcohol, peroxide are cheap.
The more you do yourself, the cheaper it is. I made a scale for $10 and cobbled some cool-white bulbs together with some ballasts.
Later you can buy extras like hormones, vitamines, pH meters, make a laminar flow hood and on and on.
If you ask around you might be able to pick up some supplies to start out with from other people on the forum. If you don't mind not being "fancy", I don't see why you couldn't start out with spending ~$75.

I had a couple of extra bottles of medium with hormones in them. They were for my abortive ceph propagation attempts. One of my VFT bottles got some mould so I transferred them to the two "hormone" bottles. They don't look so good after a few days. What looks like browning and phenolic oxidation is happening. This has never happened when transferring to non-hormone medium. All of my explant attempts have involved hormones and all died of what looked like phenolic oxidation. (The ones that didn't get contaminated).
Is it possible that hormones can cause or contribute to phenolic oxidation and explant death? It's happened with NAA, 2iP, IAA and BAP in different concentrations. Maybe I should reduce the concentrations but keep the auxin/cytokine ratios.
Any ideas from the TC crowd?
By the way, removing the seed coat from VFT seeds resulted in zero contamination in TC while the intact seeds did get some contamination.

Lloyd, how were you able to remove the vft seed coats? They are quite small.

lloyd - Might be the hormone concentration. Also, a lot of tc'ers only use hormones in the mother flask. Some use activated charcoal when adding hormones in replates. dvg - I learned this from lloyd, make sure the vft seeds are dry. Place them in a coffee filter and gently rub them together. hb

I just gently rubbed them with my finger between two pieces of paper until the seed coats flaked off.
I'll try decreasing hormone concentration plus act. char. and potassium citrate:citric acid, supposed to be antioxidants.
Thanks.

Wow, sounds like a good combination of GA3 and IBA without the elongation effect. Plus, it's soluble in water.
Are you ordering some lloyd? This will double the size of your TC specimens.

I'm thinking about it. We could split it as even 5 mg would go a LOOOOOOONG way.

Count me in lloyd. Dash3echo and myself are just waiting for the order that we did last week. Just for some culture vessels and stuff.

Lloyd, are we still doing this? I see that toptropicals are also selling this product [EPIN] but I'm still waiting for their reply. This will surely boost TC size in half the time.

Hackerberry: Yes, let's get some. Is anybody getting anything else, so we could share the S&H? I have a scale which can measure easily down to a mg. to measure it out for sharing.
